Concept
Bulk-tissue methylation clocks track how average methylation values shift with age. Single-cell techniques reveal a parallel phenomenon: variability between cells at a given locus grows with age. Cells lose epigenetic identity coherence.
Implications
- Transcriptional programs become noisier — tissues drift toward heterogeneous, sub-optimal states.
- Predicts declining function even when average values remain normal.
- Consistent with partial-reprogramming rescue: resetting epigenetic state restores identity coherence.
Measurement
- Single-cell RNA-seq and single-cell ATAC-seq quantify heterogeneity.
- Not yet a clinical biomarker; research-only.
